Calibration process - Stage

A correctly calibrated stage is precondition for being able to navigate on the sample. Carry out the Stage calibration process when you are working with low magnifications. After the calibration has been carried out successfully, you can navigate in the live-image. You can center a position on the sample by double clicking on it. Note: Calibrate the stage for every magnification that you want to work with.

If the selected magnification is too high for navigating with the stage, you receive an error message during the calibration process. Carry out the Image Shift calibration process to be able to navigate precisely at high magnifications. You can find more information on the Image Shift calibration process here.

Starting the software wizard

A software wizard will lead you through the calibration process. To start the software wizard for this calibration process, select the Stage calibration process, in the Calibrations dialog box. Then click the Calibrate... button.

Calibrating the stage

Prerequisites:

The microscope can be controlled by remote.

Valid shading correction images are available and have been activated.

The magnification has been calibrated.

Performing the calibration process

1.Open the dialog box with the Acquire > Calibrations > Stage command.

2.Click the Calibrate... button.

The Stage Calibration dialog box opens.

3.Then follow the instructions given you by the software wizard.

Set the magnification you want on the microscope.

Make sure that the sample is visible.

4.Click the Next > button.

The Stage Calibration - Check Positions dialog box opens.

The camera switches into live mode.

The stage moves to each position in turn.

You have access to the Microscope Control and Camera Control tool windows to change settings, if necessary.

5.Make sure that your sample is visible at each stage position.

6.You can interrupt the process by clicking the Pause_StageCalib button. You can continue the process by clicking the Play_StageCalib button.

7.You can abort the process at any time by clicking the Cancel button.

6.After all the stage positions have been moved to once, the Start Calibration button becomes active. You can have the stage move to each position again if you want to undertake additional settings.

7.Click the Start Calibration button when the stage positions are set correctly.

The calibration starts.

8.The Stage Calibration - Finished dialog box informs you about the success of calibration. Next to the magnifications used, the quality of the calibration is displayed.

Repeat the calibration process if the Poor entry is displayed in the Quality estimation field. To do so, click the Start Over button.

If you want to work with additional magnifications, carry out the calibration process for them. To do so, click the Start Over button.

If you want to cancel the calibration process without saving the calibration, click the Cancel button. Calibrations that are already saved are not affected by this.

9.Click the Finish button to save the calibration and to close the wizard.

10.You will then return to the Calibration dialog box. A successfully carried out calibration process is indicated for each magnification by the Calibrated status.
