Calibration process - Image Shift

The Image Shift calibration process is a prerequisite for being able to navigate precisely on the sample at high magnifications. After the calibration has been carried out successfully, you can navigate in the live-image. You can center a position on the sample by double clicking on it. You can find more information on this topic here.

Note: You have to carry out the Image Shift calibration process for every magnification that you want to work with.

If the selected magnification is too small for the image shift, you will receive an error message during the calibration process. Carry out the Stage calibration process to be able to navigate at low magnifications. You can find more information on the Stage calibration process here.

Starting the software wizard

A software wizard will lead you through the calibration process. To start the software wizard for this calibration process, select the Image Shift calibration process, in the Calibrations dialog box. Then click the Calibrate... button.

Calibrating the Image Shift

Prerequisites:

The microscope can be controlled by remote.

Valid shading correction images are available and have been activated.

The magnification has been calibrated.

Performing the calibration process

1.Open the dialog box with the Acquire > Calibrations... > Image Shift command.

2.Click the Calibrate... button.

The Image Shift Calibration dialog box opens.

3.Then follow the instructions given you by the software wizard.

Set the magnification you want on the microscope.

Make sure that the sample is visible.

4.Click the Next > button.

5.The Image Shift Calibration - Check Position dialog box opens.

6.The camera switches into live mode.

7.The preview displays the current position on the sample.

8.You have access to the Microscope Control and Camera Control tool windows to change settings, if necessary.

6.Optimize the settings in the Microscope Control and Camera Control tool windows, if necessary.

7.Click the Start Calibration button.

The calibration starts.

Note: If the current magnification is too low for the image shift, you receive an error message. In this case the calibration is canceled. Use the Stage calibration process for the current magnification.

8.The Image Shift Calibration - Finished dialog box informs you how the calibration went. Next to the magnifications used, the quality of the calibration is displayed.

Repeat the calibration process with the same magnification if the Poor entry is displayed in the Quality estimation field. Click the Start Over button.

If you want to work with additional magnifications, carry out the calibration process for them. Click the Start Over button.

If you want to cancel the calibration process without saving the calibration, click the Cancel button. Calibrations that are already saved are not affected.

9.Click the Finish button to save the calibration and to close the wizard.

You will then return to the Calibration dialog box. A successfully carried out calibration process is indicated for each magnification by the Calibrated status.
