Use this dialog box to select settings for the autofocus.

You open this dialog box via the Camera Control tool window. In the tool window's toolbar, click the Acquisition Settings button. Select the Acquisition > Autofocus option in the tree view.

Maximum Z-range

In the Maximum Z-range field, you define the Z-range the software will consider during the Autofocus calibration calibration process in order to acquire images for the calculation of the focal point. By default, a value of 5000 nm is defined. Enter a value into the field or use the arrow buttons to increase or decrease it.

Reduce the value in the Maximum Z-range field if the Microscope Alignment calibration process cannot be carried out successfully. By doing so, you reduce the image displacement during the calibration process.

Autofocus loops

In the Autofocus loops field, you set the number of repetitions that will be used in order to find an optimal focus position. By default, 2 loops are set. Several repetitions can improve the accuracy of the Autofocus process. However, the duration of the process will increase by them. Enter a value into the Autofocus loops field or use the arrow buttons to increase or decrease the number of loops.

Magnification range and User Defocus (nm)

In the Magnification range column, you assign a defocus to every magnification range of your microscope. You can also split the magnification range and define several magnification ranges to which you assign an individual defocus. When you carry out your software's Autofocus function, the selected magnification range's defocus is automatically subtracted from the determined Gaussian focus. In certain cases, a defocus is helpful, for example for adjusting the image contrast individually.

If you want to split a magnification range, select this magnification range in the table and click the Split... button. The Split magnification range dialog box opens. Select a magnification you want to split the selected magnification range with from the list and click OK. The newly created magnification range is added to the table. It has the same defocus than the previously split magnification range. You can change the defocus in the User Defocus (nm) column. If you defined more than one magnification range, you can delete them with the Delete button. If you delete a magnification range, the next higher magnification range will be used.

Clicking the Default button will reset the Maximum Z-range and Autofocus loops fields to their default values.
