The Focus Indicator toolbar is there for you to use when you are focusing on your sample. Note: You can only make the Focus Indicator toolbar appear in the live-image. Additionally, you can use the Line Profile tool window to check the focusing. Switching the focus indicator on or offClick this button Alternatively, you can also use the View > Toolbars > Focus Indicator command. Structure of the Focus IndicatorThe Focus Indicator consists of two bars. The red bar shows the sharpest focusing that has already been achieved. The blue bar shows the sharpness of focus that is being achieved with the current focusing. The blue bar will be continuously updated and varies between "blurred" (left) and "focussed" (right).
Resetting the Focus IndicatorWhen the Focus Indicator's bar has already reached the right-hand edge, although the image is still very blurred, click the Reset Focus Indicator The Reset Focus Indicator function can also be used if the sharpness drops very drastically due to changes made to the microscope settings while an acquisition is in progress. Focusing with the focus indicator1.Go to the place in the sample that interests you and switch to the live mode. 2.Should the Focus Indicator not be visible, switch it on by clicking this button 3.A rectangle will appear in the image. The maximum contrast serves as a monitor for the sharpness in this area. 3.Change the rectangle's position and size, to have an area of the image that especially interests you measured. Make sure that the area within the rectangle contains a part of the sample with plenty of contrast. 4.Adjust the focus settings on your microscope. While doing this, watch the bar in the Focus Indicator. 5.The red bar shows the greatest sharpness that has up till then been achieved. 6.The blue bar shows the sharpness of focus that is being achieved with the current focusing. 5.Change the focusing on your microscope until the display is as completely blue as possible. 6101 26032013 |
